Is SAMBA 4 out?
I can tell you from previous experience with samba 3 that small installations are not bad. Large installations would be a real pain in the you know what. Managing users from LDAP is the way to go. Assuming one can configure LDAP.
SAMBA 4 was supposed to have LDAP as part of the install. I would like to see it in action.

Samba 4 is a powerful answer to your question.. I don't have any
direct experience, however hanging out in the IRC channels from time
to time you tend to see the phrase "ADS eh?, you wan't Samba4..."
quite a bit.
